alt-enter should work just fine to switch it fullscreen. There's also an option you can set to always have it fullscreen I think, but I use D-Fend, so I don't remember how to do it anymore.

Change the line

fullscreen=false

to

fullscreen=true

and set the resolution in the config file. or alt-enter. It's clearly listed in the readme and in the site's DOSBox newbie's guide.

If you want the resoulution to fill out your new full screen, you can use full=fixed in a line right next to it. Or something like that.

Would the game run slower in full-screen mode?

No, it should run way smoother.
